Transient receptor potential ion channels serve as polymodal-regulated sensors, which are widely expressed in diverse tissues of higher organisms. A subset of the TRP channel family senses biological temperature, where changes in thermal energy are converted to conformational change (work), a feature central to thermosensing and thermoregulation. In addition to functioning as molecular thermometers, these proteins are regulated by a variety of other stimuli including modulatory proteins, voltage, pH, chemical ligands and lipids. This seminar will detail efforts to untangle and understand the molecular mechanisms of TRP channel stimulatory integration and regulatory modes through structural, functional and thermodynamic approaches. The data will focus on Van Horn’s studies of TRPV1 and TRPM8 ion channels and the emerging view of the mechanistic interplay in TRP channel structure and function.
